Modified method for obtaining phycocyanine concentrate of Arthrospira platensis biomass

Abstract

The biomass of Arthrospira platensis cyanobacteria is a source of bioactive compounds such as chlorophylls, carotenoids, and, particularly, phycobiliproteins: C-phycocyanin and allophycocyanin. The wide range of biological activity shown by extracts with a high content of phycocyanins determines the prospects for their use as dietary supplements and ingredients of special foods. For food purposes, the degree of purity of phycocyanin concentrates, determined by the ratio of optical densities of their aqueous solutions at two wavelengths, namely D620/D280, must be greater than 0.7. Most methods for obtaining phycocyanin concentrates include laborious steps of fractional ammonium sulphate precipitation of protein from A. platensis biomass extracts followed by removal of salts solution. The use of membrane technology, specifically microfiltration, makes it possible to significantly intensify and simplify the process of obtaining phycocyanin concentrates.

The aim of this research was to modify the method for obtaining a high-purity A. platensis phycocyanin concentrate by replacing the stage of ammonium sulfate precipitation of the protein by ultrafiltration of the extract followed by microfiltration.

Material and methods. A sample of dry A. platensis biomass was used as a feedstock. Extraction of A. platensis biomass was carried out at a temperature of +40 °C for 3 h, the resulting suspension was centrifuged, and the supernatant was separated from the sediment. The obtained extract was subjected to ultrafiltration (membrane with a pore diameter of 30 kDa) followed by removal of the permeate containing low molecular weight impurities. The retentate was subjected to microfiltration (membrane with a pore size of 0.2 μm), concentrated by reverse osmosis and freeze-dried.

Results. The content of C-phycocyanin and allophycocyanin in the dry concentrate was 42.0±1.3 and 7.0±0.3%, respectively, the degree of purity was 1.98.

Conclusion. The scheme for obtaining A. platensis phycocyanin concentrate has been modified. A concentrate was obtained with a high degree of purity, allowing its use in food.

Каждый эксперимент был выполнен в 8 повторностях. Статистическую обработку данных проводили с использованием программного пакета IBM SPSS Statistics 20 (IBM, США). Для всех показателей, представленных в таблице, вычисляли среднее значение (М) и стандартную ошибку среднего (m). Статистические различия оценивали с использованием критерия Стьюдента. Критический уровень значимости нулевой статистической гипотезы (p) принимали равным 0,05.

Результаты и обсуждение

Белково-пептидные профили промежуточных продуктов и концентрата 2, полученных по предложенной модифицированной схеме (рис. 2А-В), и концентрата 1, полученного по технологии, включавшей стадию сульфатно-аммонийного осаждения белка из экстракта А. platensis (рис. 2Г), представлены соответствующими хроматограммами. Сравнительный анализ хроматограмм на рис. 2А и Б показал, что в результате ультрафильтрации экстракта происходит частичная элиминация балластных низкомолекулярных фракций (<12 кДа) и их содержание в отобранном ретентате снижается в 5,7 раза. Результаты, представленные в таблице, свидетельствуют о том, что при использовании модифицированной схемы в ретентате содержание белка повышается в 1,7 раза, а суммарное содержание фикоцианинов - в 2,8 раза. Последующая микрофильтрация ретентата практически полностью удалила высокомолекулярные примеси из его состава (см. рис. 2Б и В), дополнительно повысила содержание общего белка и суммарное содержание фикоцианинов в конечном продукте соответственно в 1,2 и 1,6 раза. Степень чистоты ретентата в результате микрофильтрации увеличилась в 1,8 раза. В таблице приведены данные по сравнительной характеристике концентратов 2 и 1. Суммарное содержание фикоцианинов в концентрате 2 в 1,2 раза выше по сравнению с концентратом 1. В 1,8 раза повышена чистота концентрата 2, при получении которого была исключена трудоемкая стадия сульфатно-аммонийного осаждения.

Таким образом, полученные результаты подтвердили эффективность предложенной схемы модификации процесса. Сульфатно-аммонийное осаждение белков из экстрактов А. platensis позволяет примерно в 2 раза повысить на этой стадии степень чистоты получаемого промежуточного продукта [21, 23, 24]. Последующая ультрафильтрация, как при получении концентрата 1, используется, чтобы перевести осажденный белок в растворимое состояние, удаляя сульфат аммония из раствора. В модифицированной схеме ультрафильтрация непосредственно экстракта А. platensis заменяет стадию сульфатно-аммонийного осаждения, в 1,8 раза повышая чистоту продукта. Последующая микрофильтрация, как отмечено выше, дополнительно еще в 1,8 раза повышает чистоту концентрата 2.

Заключение

Модифицирован и апробирован способ концентрирования и очистки фикоцианинов из биомассы А. platensis. Получен концентрат с высоким содержанием фикоцианинов и степенью чистоты, позволяющей использовать его в составе пищевой продукции.
