Influence of the rat diet enrichment with oat β-gucans on the assimilation of B group vitamins, mineral elements and lipid metabolism
AbstractDespite the widespread use of oat β-glucans as ingredient of foods and dietary supplements, there is insufficient data on their effect on the metabolism of vitamins and minerals.
The purpose of the study was to evaluate the effect of including oat bran with a high content of β-glucans (β-glucan) in the diet on the absorption of micronutrients and lipid metabolism in growing rats deficient in vitamins D, group B and trace elements (iron, copper, zinc).
Material and methods. After the development of micronutrient deficiency (for 23 days), in order to assess the effect of oat bran (5%) with a high content of β-glucans on the correction of the micronutrient status of growing male Wistar rats (with initial body weight of 70.7±0.7 g), the missing micronutrients were introduced in the semi-synthetic diet deficient in vitamins D, group B, iron, copper and zinc within 7 days either along with β-glucan (1.47%) or without its addition. Indicators of micronutrient sufficiency (riboflavin serum concentration, daily urinary excretion of thiamine, riboflavin and 4-pyridoxic acid, measured by fluorometric methods; serum concentration and urinary excretion of calcium, magnesium, iron, zinc, copper, phosphorus, measured by the atomic absorption method or using standard methods on a biochemical analyzer) and the biochemical parameters of blood serum were compared with the parameters of rats adequately provided with all micronutrients throughout the experiment.
Results. Replenishment of missing micronutrients in the diet of rats with deficiency in vitamins D and group B, iron, copper and zinc for 7 days led to the elimination of deficiency of vitamins B1, B2 and B6, regardless of the presence of β-glucans in the diet. At the same time, against the background of the presence of β-glucans in the feed, an increase in the absorption of iron was observed, as evidenced by an increase by 1.73 times in iron blood plasma level (р<0.05) and a tendency towards its urinary excretion decrease by 1.60 fold (р<0.10) compared to animals from the control group. Adding oat bran with β-glucans to the feed did not lead to a decrease in blood plasma level of total cholesterol and low-density lipoproteins cholesterol. The levels of high-density lipoprotein cholesterol and triglycerides in rats of all three groups did not have statistically significant differences.
Conclusion. The presence of β-glucans in the diet had virtually no effect on the absorption of B vitamins and improved the absorption of iron.

Через 7 сут коррекции микронутриентного статуса крыс уровень в сыворотке крови ХС ЛПНП, повышающийся при дефиците витаминов и, в частности, при недостатке витамина D [26-28], остался повышенным в группах крыс, получавших рацион как с добавлением овсяных отрубей с β-глюканами (группа "+D+B+Ме+β-глюкан"), так и без их добавления ("+D+B+Ме") в 1,4 и 1,5 раза по сравнению с показателем крыс контрольной группы (рис. 2), не прошедшей стадию дефицита микронутриентов.
Иными словами, добавление в корм овсяных отрубей с β-глюканами в течение 1 нед не привело к снижению уровня ОХС и ХС ЛПНП в сыворотке крови. Уровень липопротеинов высокой плотности, глюкозы, триглицеридов у крыс всех 3 групп не имел статистически значимых различий.
Заключение
В современной литературе в исследованиях, посвященных изучению влияния на липидный обмен и чувство насыщения, β-глюканы овса относят к растворимым пищевым волокнам [3, 29], в исследованиях, направленных на изучение влияния на микробиоту кишечника и иммунную систему, их рассматривают в качестве пребиотиков [1, 8, 30]. В технологических целях при производстве пищевых продуктов их используют в качестве загустителей, стабилизаторов и заменителей жира [1].
Для исследования влияния овсяных отрубей с повышенным содержанием β-глюканов на усвоение микронутриентов у растущих крыс на первом этапе был создан дефицит микронутриентов (витаминов D, группы В, железа, меди и цинка) путем уменьшения в корме соответствующих пищевых веществ, а затем содержание недостающих микронутриентов в корме животных было доведено до адекватного уровня либо на фоне включения в рацион овсяных отрубей с высоким содержанием β-глюканов, либо без их добавления. Предполагалось, что сравнение содержания витаминов и макро- и микроэлементов в сыворотке крови и моче в конце эксперимента позволит выявить возможное влияние пищевых волокон и β-глюканов на усвоение микронутриентов.
Полученные результаты свидетельствуют о том, что обогащение рациона овсяными отрубями с высоким содержанием β-глюканов не оказывает заметного влияния на усвоение витаминов группы В (В1, В2 и В6) и повышает усвоение железа.
В настоящее время пребиотики рассматривают в качестве альтернативного и эффективного метода повышения усвояемости кальция и минеральной плотности костной ткани у лиц с недостаточным потреблением кальция. Пищевые волокна (пребиотики) усиливают усвоение минеральных веществ и могут способствовать здоровью костей, как это было продемонстрировано на модели постменопаузы у овариэктомированных крыс [10]. Подтвердить улучшение усвоения кальция и магния в данном исследовании не удалось, так как измерения этих макроэлементов производили в сыворотке крови, концентрация в которой поддерживается на постоянном уровне и не в полной мере отражает их внутриклеточное содержание [2, 31]. Вместе с тем полученные в данном эксперименте результаты не отвергают возможности улучшения биодоступности этих макроэлементов, доказанного в других исследованиях путем их измерения в костной ткани [8, 10].
В нашем эксперименте в качестве источника β-глюканов были использованы овсяные отруби с высоким содержанием β-глюканов (29,4%). Для большинства коммерчески доступных ингредиентов с повышенным содержанием β-глюканов изготовителем обычно гарантированы такие показатели, как вязкость и процентное содержание пищевых волокон, но без указания их молекулярной массы, однако, учитывая зависящее от молекулярной массы меньшее влияние на регуляцию уровня ХС и гликемический контроль, многие авторы пришли к выводу, что такая информация необходима [17], поскольку использование β-глюканов с разными физико-химическими свойствами может приводить к получению противоречивых результатов. Отсутствие гипохолестеринемического и гипогликемического действия овсяных отрубей с высоким содержанием β-глюканов у крыс, получающих стандартный по содержанию жира и углеводов рацион при дефиците витаминов и некоторых минеральных веществ в условиях нашего эксперимента, может указывать на преимущественное наличие в используемом образце овсяных отрубей β-глюканов с низкой молекулярной массой.
Результаты исследования свидетельствуют о целесообразности включения в состав эффективного для коррекции минерального статуса пищевого продукта или биологически активной добавки к пище, наряду с минеральными веществами, β-глюканов.
