Influence of chronic immobilization stress on vitamin status in rats fed different diets

Abstract

The influence of a stress factor, widespread in modern conditions, on the vitamin status has not been studied enough. At the same time, the negative stress impact can be aggravated against the background of unhealthy nutrition, which in turn affects the vitamin status of the organism.

In this regard, the goal of the research was to evaluate the effect of chronic restrict stress on the vitamin supply in rats fed a diet with adequate and increased content of fat, sugar and cholesterol.

Material and methods. The experiment was carried out on 37 growing male Wistar rats (initial body weight of 45±5 g) divided into 4 groups. Animals of the 1st (control) and the 2nd groups received a complete semi-synthetic diet (CSSD) (20% protein, 10% fat, 58% carbohydrates in the form of starch, 384 kcal/100 g) for 92 days. The levels of all vitamins and mineral elements in the rats’ diets were adequate for growing rats. Rats of the 3rd and the 4th groups were fed a high-calorie, high-fat high-carbohydrate diet (HFHCD) (20% protein, 28% fat, 2% cholesterol, 18% carbohydrates in the form of starch, 20% sucrose, 511 kcal/100 g). Animals of groups 2 and 4 were subjected to daily 90-minute immobilization. The concentration of vitamins A (retinol and retinol palmitate) and E (α-tocopherol) in the blood serum and liver were determined by high-performance liquid chromatography, vitamins B1 and B2 in the liver and urine, as well as riboflavin in the blood serum and 4-pyridoxic acid (4-PA) in urine were determined by fluorimetric methods. Biochemical parameters of blood serum were determined on a biochemical analyzer; the total content of fat, triglycerides (TG) and cholesterol (CH) was determined in the liver.

Results. Replacing CSSD with HFHCD, both under restraint stress and without, was accompanied by an increase in liver weight by 1.8–2.0 fold, in its fat content by 2.6–3.3 fold, cholesterol by 32.6–35.3 fold and TG – by 33.0–57.6 fold (p≤0.001). An increase in alanine aminotransferase (ALT) activity by 1.7–2.0 fold (p≤0.01), in low-density lipoprotein (LDL) cholesterol level by 5.4 fold (p≤0.05) and the atherogenic coefficient by 2.5 fold (p<0.01) as well as a decrease in creatinine and urea level (p≤0.05) in blood serum were revealed. Immobilization was accompanied by a decrease in body weight, liver and liver fat in rats fed both CSSD and HFHCD (p<0.05), but didn’t affect the blood serum biochemical parameters, with the exception of an increase in ALT activity. If the activity of alkaline phosphatase (ALP) did not change during immobilization of rats fed the CSSD, then in animals fed the high-calorie diet it decreased by 37.5% (p≤0.05 from the control) under its increase against the background of restrict stress by 78.7% (p≤0.01) compared to the indicator of rats of the 3rd group. Immobilization of rats treated with CSSD was accompanied by an increase in both absolute serum α-tocopherol level and concentration correlated with the level of cholesterol and triglycerides by 26.0–57.5% (p<0.05), with a simultaneous decrease in its content in the liver per 1 g of wet tissue by 22.1% (p=0.041) relative to the indicators of intact animals. Immobilization reduced the level of retinol palmitate in the liver by 2.3 times (p<0.01), but did not affect retinol level in the blood serum. At the same time, indicators of B vitamin status (the content of vitamins B1 and B2 in the liver per 1 g of wet tissue and per organ, blood serum riboflavin level, urinary excretion of riboflavin and 4-PA) did not change, with the exception of thiamine urinary excretion, which reduced compared to the control by 38.8%. In rats fed HFHCD, immobilization had no additional effect on the supply with vitamins A and E. The content of vitamins B1 and B2 in the liver in terms of the whole organ was reduced by 14.0–26.7% relative to the indicator in animals of the 3rd group, not subjected to chronic stress, only due to differences in liver weight in animals of these groups.

Conclusion. The data obtained indicate that chronic stress has a negative effect on the vitamin status of the body, worsening the supply with vitamins A, E and B1, and substantiate the feasibility of studying the mechanisms of this effect in order to develop effective vitamin complexes for the treatment and prevention of diseases caused by long-term stress.

Хронический иммобилизационный стресс на фоне ППСР сопровождался некоторым ухудшением обеспеченности витамином В1, о чем свидетельствовало снижение суточного выведения этого витамина с мочой. Кроме того, запасы витамина А в печени стрессированных крыс уменьшились более чем в 2 раза при сохранившейся на неизменном уровне концентрации ретинола в сыворотке крови. Последнее согласуется как с известным механизмом гомеостатического поддержания постоянной концентрации в крови за счет расхода витамина А, депонированного в печени, так и с данными некоторых авторов об уменьшении содержания витамина А в печени крыс, подвергавшихся хронической иммобилизации. Так, отмечалось, что ежедневная 4-часовая иммобилизация крыс в течение 7-14 сут вызывала изменение в накоплении в тканях витамина А: его содержание в печени, семенниках и почках и в сыворотке крови уменьшалось, а в надпочечниках постепенно увеличивалось, достигая максимума на 7-е сутки [26]. Однако, по данным других авторов, в 10-дневном эксперименте стрессовое состояние, вызванное иммобилизацией, приводило к накоплению ретинилпальмитата в печени [27].

Регулярная иммобилизация крыс, получавших стандартный рацион, приводила к повышению как абсолютной, так и соотнесенной с ХС и ТГ концентрации витамина Е в сыворотке крови за счет снижения его содержания в печени. По всей видимости, рост концентрации α-токоферола в крови обусловлен компенсаторной необходимостью предотвращения нарастания интенсивности ПОЛ при стрессе. Аналогичное уменьшение уровня витамина Е в печени на фоне выраженного нарастания концентрации продуктов ПОЛ в крови и печени наблюдалось после водно-иммерсионного стресса (WIRS) у крыс, содержавшихся в течение 4 нед, но на витамин Е-дефицитном рационе [28].

По данным литературы, при остром иммобилизационном стрессе происходит перераспределение пула витамина Е у крыс. Так, однократное 6-часовое воздействие WIRS на крыс оказывало разнонаправленное действие на уровень витамина Е в различных органах. Оно не оказывало влияния на концентрацию его в сыворотке крови, но снижало содержание витамина Е в тимусе на 31% и повышало в селезенке на 40% при наблюдающемся увеличении концентрации аскорбиновой кислоты в сыворотке крови, по-видимому, вследствие усиленного ее высвобождения из поврежденной печени [29]. В другом исследовании этих же авторов было показано повышение концентрации витамина Е в почках и сердце, но не в печени и снижение в скелетных мышцах [30]. При этом предварительное пероральное введение витамина Е (50 или 250 мг/кг массы тела) хотя и не полностью, но предотвращало это снижение.

В то же время одновременная экспозиция крыс холодовому и иммобилизационному стрессу (1 ч при температуре +4 °С 5 дней в неделю), но в течение более длительного времени (21 сут) не оказывала влияния на уровень витамина Е в печени, сердце и мозге, однако приводила к снижению содержания витамина С в печени и головном мозге [31].

При сравнении показателей крыс 3-й и 4-й групп оказалось, что иммобилизация крыс на фоне ВЖВУР не оказала значимого влияния на обеспеченность витаминами А и Е. Общие запасы витаминов В1 и В2 в печени оказались сниженными на 14,0-26,7% относительно показателя животных 3-й группы, не подвергавшихся хронической иммобилизации, только вследствие существенно меньшей массы печени.

Увеличение содержания жира в 3 раза, ХС и углеводов в рационе не сопровождалось изменением концентрации рибофлавина, ретинола и α-токоферола в сыворотке крови и суммарного содержания витамина А в печени. В то же время потребление такого рациона привело к существенному накоплению витамина Е в печени, сопряженного с увеличением ее массы и жирового депо. Аналогичное повышение уровня α-токоферола в печени (на 46-89%, р<0,05) при отсутствии изменения концентрации α-токоферола в плазме крови было обнаружено у крыс при увеличении содержания жира в рационе с адекватного до 30-31% [18, 32]. При этом в данном исследовании вследствие избыточного накопления липидов в печени соотнесенное с ними содержание этого витамина снизилось практически на порядок, как и соотнесенная с ХС ЛПНП концентрация α-токоферола в сыворотке крови, что может свидетельствовать об ухудшении антиоксидантного статуса организма.

Обнаруженное увеличение общих запасов витамина В2 в печени при замене ППСР на ВЖВУР согласуется с ранее полученными нами данными у крыс при увеличении содержания жира в рационе до 30% [18], однако оно наблюдалось при снижении удельного содержания этого витамина в органе.

В то же время замена ППСР на ВЖВУР на фоне хронической иммобилизации, в отличие от нестрессированных животных, приводила к тому, что общие запасы витамина А в печени были выше, чем у иммобилизованных животных с адекватным поступлением жира и углеводов, и не отличались от контроля. Это, по-видимому, свидетельствует о компенсации снижения запасов витамина А при стрессе за счет ВЖВУР. Отсутствие изменения абсолютной и соотнесенной с ХС концентрации α-токоферола в сыворотке крови у крыс на ВЖВУР при стрессорном воздействии привело к тому, что на фоне иммобилизации увеличение содержания жира и углеводов в рационе этот показатель обеспеченности витамином Е был ниже, чем у получавших ППСР животных. Данный результат, по-видимому, следует рассматривать как неблагоприятный.

В целом анализ воздействия 2 факторов свидетельствует, что наиболее значимое влияние на показатели обеспеченности витаминами оказало изменение рациона, а также иммобилизация на фоне потребления ППСР.

Заключение

Проведена оценка влияния хронического иммобилизационного стресса в сочетании с высококалорийным рационом на статус водо- и жирорастворимых витаминов в организме. Особый интерес представляют полученные данные о снижении в этих условиях показателей обеспеченности витамином Е, что свидетельствует о развитии системного окислительного стресса, как возможного звена патогенетического механизма метаболического синдрома при стрессе.

Результаты проведенного исследования свидетельствуют, что хронический стресс у крыс, получавших ППСР, оказывает негативное влияние на витаминный статус организма, ухудшая обеспеченность витаминами А, Е и В1. Несмотря на то что потребление ВЖВУР несколько смягчало это воздействие, необходима коррекция обеспеченности витаминами организма, находящегося в состоянии хронического стресса. Полученные данные обосновывают целесообразность изучения механизмов воздействия стресса на витаминный статус и последующей разработки перспективных витаминных комплексов для лечения и профилактики заболеваний, вызванных длительным стрессом.
