Alternaria toxins in tomato products marketed in the Russian Federation

Abstract

Tomatoes and tomato products are widely produced and consumed throughout the world. Alternaria spp. are the main cause of alternariosis (black mold disease) on fresh tomatoes, both in the field and after harvesting. Alternaria toxins are widespread contaminants of tomato products.

The aim of the present study was to evaluate the contamination of tomato processing products from the domestic market with Alternaria toxins, as well as to assess their intake by humans through the consumption of tomato juices.

Material and methods. The content of Alternaria toxins (alternatiol, alternariol monomethyl ether, altenuene, tentoxin, tenuazonic acid) was determined in 64 samples of tomato products (paste, ketchup, juice) by high-performance liquid chromatography coupled to tandem mass-spectrometric detection (HPLC-MS/MS).

Results. The priority Alternaria toxins for tomato paste, ketchup and juice were tenuazonic acid (61% of 64 samples, in amounts from 20.0 to 1065.5 μg/kg), altenuene (52%, 8.9–200.1 μg/kg) and alternariol (27%, 12.2–561.6 μg/kg). Samples of tomato paste turned out to be the most contaminated with Alternaria toxins while tomato juice samples were the least contaminated. At the same time, several toxins were found in 91% of tomato paste samples, 35% of ketchups, and 23% of tomato juices.

Conclusion. To the best of our knowledge, the present study is the first survey devoted to Alternaria toxins contamination of tomato paste, ketchup and tomato juice sold on the Russian market. The high frequency of their contamination with tenuazonic acid, altenuene and, to a lesser extent, alternariol has been established, which indicates a potential risk to human health when tomato processing products are consumed. This indicates the need for a hygienic assessment of contamination the above products with tenuazonic acid, altenuene and alternariol. When calculating the potential intakeof Alternaria toxins for different age population groups, it was shown that high levels of alternariol (up to 56.77 ng/kg body weight per day) could be obtained under daily consumption of tomato juice by adults and children under three years of age, as well as tenuazonic acid when consuming tomato juice contaminated at the 95th percentile level as part of the diet in organized groups for orphans and children without parental care.

Микотоксины - токсичные метаболиты плесневых грибов - наиболее опасные природные загрязнители продовольственного сырья. Контроль загрязнения ими сельскохозяйственной продукции является условием предупреждения недопустимого риска, связанного с вредным воздействием на человека и будущие поколения. Наиболее распространенными микотоксинами являются токсины грибов рода Fusarium, Alternaria, Aspergillus и Penicillium.

Помидоры и продукты их переработки широко производятся и потребляются во всем мире. В 2020 г. производство томатов занимало 1-е место среди овощных культур в мире [1]. По данным Минсельхоза России, урожай овощей в круглогодичных теплицах в 2022 г. значительно увеличился [2]. Около 70% продукции томатов предназначено для переработки на пюре, мякоть, соусы, соки и кетчупы.

Кетчуп является вторым по популярности соусом в России после майонеза. В 2022 г. российскими предприятиями было выпущено кетчупа и томатных соусов на 12,1% больше по сравнению с 2021 г. Потребление кетчупа занимает почти 13% от объема всех соусов и специй, продаваемых в России [3].

Известно, что микроскопические грибы Alternaria spp. являются основной причиной альтернариоза (сухая пятнистость) на свежих томатах, как в поле, так и после сбора урожая [4-6]. По нашим данным, в свежих томатах обнаруживали тентоксин (TEN, частота обнаружения - 27%), альтернариол (АОН, 9%) и тенуазоновую кислоту (ТеА, 9%); в плесневелых томатах частота загрязнения и уровни альтернариатоксинов значительно выше [7].

Установлена загрязненность микотоксинами Alternaria (альтернариатоксины, АТ) различных томатных продуктов [8-11]. По данным Европейского агентства по безопасности пищи (EFSA), АОН и ТеА наиболее распространены среди АТ. Их обнаруживали чаще в зерновых, томатах и продуктах их переработки [12, 13].

В тесте Эймса и на культурах клеток млекопитающих (в экспериментах in vitro) было установлено, что AOH и метиловый эфир альтернариола (AME) обладают мутагенными и генотоксическими свойствами. Получены единичные данные о степени токсичности АТ в экспериментах in vivo. Так, LD50 при пероральном введении мышам и крысам натриевой соли ТеА составила 81-186 мг/кг массы тела в сутки. Таким образом, данное вещество относится к веществам 3-го класса опасности [14, 15]. Употребление загрязненной АТ пищи рассматривают как возможную причину высокой заболеваемости раком пищевода в провинции Хэнань (Китай) [16, 17]. Высокую степень загрязненности пищевых продуктов ТеА связывают с развитием тяжелого заболевания Оньялаи (onyalai; purpura thrombopenica tropica acuta; острая тропическая тромбоцитопеническая пурпура), встречающегося в странах Африки к югу от пустыни Сахара [18].

В Российской Федерации ранее не изучали загрязненность токсинами грибов рода Alternaria продуктов переработки томатов. Учитывая, что, по данным EFSA, томаты и продукты из них являются одними из основных источников воздействия АТ с пищей на человека, целью данной работы было изучение частоты и уровня загрязнения ими томатной пасты, кетчупа и томатных соков, реализуемых на территории России, и провизорная оценка потенциального риска здоровью для населения России.

Материал и методы

Образцы продуктов переработки томатов покупали на предприятиях розничной торговли Москвы и Московской области случайным образом. Исследовано 64 пробы продукции: 22 образца томатной пасты, 20 образцов кетчупа и 22 образца томатного сока отечественного и импортного производства.

Подготовку образцов для скрининга АТ в продуктах из томатов проводили в соответствии с разработанной методикой. В пробирку типа фалькон объемом 50 см3 отбирали измельченную пробу массой 5 г, добавляли 5 см3 дистиллированной воды (только для кетчупа и томатной пасты) и перемешивали, добавляли 10 см3 ацетонитрила, подкисленного 1% муравьиной кислотой, после 1 мин перемешивания на вортексе встряхивали на шейкере в течение 10 мин. Добавляли 1 г NaCl и 4 г безводного MgSO4, интенсивно перемешивая вручную или на вортексе. Экстракт центрифугировали в течение 5 мин при 7000 об/мин на центрифуге Rotina 38 (Hettich, Германия), затем отбирали 1 см3 супернатанта и упаривали досуха на роторном испарителе (Rotovac, Германия). После этого перерастворяли в 500 мм3 смеси ацетонитрил : вода (20:80 об.). Полученный раствор переносили в пробирку типа эппендорф объемом 1,5 см3 и центрифугировали в течение 10 мин при 15 000 об/мин на центрифуге (Thermo Scientific, США). В хроматографическую виалу вносили 100 мм3 супернатанта, после добавления 100 мм3 метанола проводили идентификацию и количественное определение АТ методом обращенно-фазовой высокоэффективной жидкостной хроматографии высокого давления с тандемным масс-спектрометрическим детектированием (ВЭЖХ-МС/МС) в режиме положительной электрораспылительной ионизации при атмосферном давлении и динамического мониторинга выбранных переходов.

Анализ АТ проводили с использованием ВЭЖХ-системы (Agilent Technologies 1100), состоящей из градиентного насоса, термостата колонок, автосамплера и соединенной с тройным квадрупольным масс-спектрометрическим детектором (Triple Quad 6400). Разделение аналитов осуществляли на колонке, заполненной силикагелем с привитыми группами октадецилсилана (Zorbax SB-С18, 150×4,6 мм, 3,5 мкм, размер пор - 80Å, Agilent), в режиме градиентного элюирования смесью вода : ацетонитрил, модифицированной 0,1% муравьиной кислотой.

Использованы стандарты: AOH (99,3%), AME (99,77%), альтенуен (ALT) (98%), тентоксин (ТЕN) (99,84%) (Fermentek, Израиль). Исходные растворы этих токсинов готовили в метаноле в концентрации 200 мкг/cм3.

Стандарт ТеА с концентрацией 100,6 мкг/cм3 был получен из компании
Romer (Romer Labs, Biopure, Австрия). Все исходные растворы хранили при -18 °C.

Матричный эффект для всех токсинов, кроме ТеА, был незначительным и не превышал 18%, для ТеА составил 30%. Степени извлечения токсинов варьировали от 86% (ТеА) до 110% (АМЕ). Пределы количественного определения метода для TEN, AME, AOH, ALT и TeA составили 1, 4, 6, 8 и 20 мкг/кг соответственно.

Оценку риска здоровью, обусловленного загрязненностью АТ томатного сока, проводили на основании сравнения величины суточного поступления АТ с величиной суточного порога их токсического воздействия (ТТС) на человека. Расчетное суточное поступление токсинов с томатным соком рассчитывали по формуле:
