Contractive function of mesenteric lymph nodes in obese rats

Abstract

Over the past 50 years, the prevalence of obesity around the world has increased several times and has become a pandemic. The effect of obesity on the lymphatic system, which plays a key role in the regulation of fluid homeostasis, immune cell migration, antigen presentation, and resolution of inflammatory responses, is poorly understood, and there is no data on the contractile activity of the lymph nodes in obesity.

The purpose of the research was to investigate the parameters and mechanisms of dysfunction of the contractile function of the mesenteric lymph nodes of rats in obesity caused by the feeding with the high-fat diet (HFD).

Material and methods. The study was conducted on 50 male Sprague-Dawley rats. Rats aged 6 weeks were randomly divided into groups: a control group (n=10) fed a standard diet and a group of rats (n=40) kept on HFD (60% fat content by calorie value). Rats received food and water ad libitum for 16 weeks. Before the end of the experiment, four groups of HFD rats were formed: obesity resistant animals (HFD-OR, n=11), without additional interventions (HFD, n=10), rats which were administered dexamethasone three days before the study (HFD+Dexa, n=9), HFD followed by 8-week diet restriction (HFD+DR, n=9). At the end of the experiment, mesenteric lymph nodes (LNs) were taken from rats under anesthesia and their contractile function was studied in a myograph using 1400W, dynastat and Tempol.

Results. LNs of control rats had a high level of tone and generated spontaneous high-amplitude phasic contractions. The LNs of HFD rats had a low initial tone, and rare low-amplitude phasic contractions were recorded in them. The parameters of contractile activity of the LNs of rats in HFD-OR and HFD+Dexa groups differed slightly from the corresponding parameters of the LNs of rats in the control group. Calorie restriction for 8 weeks in obese rats (HFD+DR) resulted in an increase in tone, frequency and amplitude of phasic contractions of the LNs compared to those in HFD rats. iNOS inhibition caused a significant increase in the tone, amplitude and frequency of phasic contractions of the LNs in the HFD group. An increase in the frequency of phasic contractions was observed only in the LNs of HFD+Dex and HFD+DR rats. Inhibition of cyclooxygenase 2 did not affect the contractile function of the LNs of rats of all groups, with the exception of animals from the HFD group (increase in the amplitude and frequency of phasic contractions). Tempol significantly increased the tone, frequency and amplitude of phasic contractions of the LNs in rats of the HFD group and increased the frequency of phasic contractions of the LNs of the HFD+DR rats.

Conclusion. A high-fat diet leads to impaired contractile function of rat LNs and can create additional obstacles to the movement of lymph, promoting its leakage into surrounding tissues. Obesity is accompanied by the development of inflammation in the LNs and perinodal adipose tissue, which induces the expression of inducible NO synthase, cyclooxygenase-2 and the accumulation of reactive oxygen species (ROS). NO, prostaglandins and ROS have an inhibitory effect on the SMC capsules of the LNs, leading to a decrease in tonic tension and a weakening of spontaneous phasic contractions. The reason for inhibition of LN contractile function is obesity, but not consumption of food high in fat. Transferring obese rats to a calorie-restricted diet results in a decrease in body weight and visceral fat mass and an improvement in LN contractile function.

Известно, что ключевым механизмом, который регулирует при ожирении патофизиологические изменения в различных системах органов, является хроническое вялотекущее воспаление [23], которое в лимфатической системе способствует перилимфатическому накоплению воспалительных клеток [6] и снижению сократительной способности ЛС [10]. Исходя из этих представлений, крысам одной из групп, получавших ВЖР, за 3 дня до исследования был введен дексаметазон, оказывающий выраженное противовоспалительное действие. ЛУ этих крыс демонстрировали значительно лучшие показатели сократительной функции: достоверное повышение тонуса ЛУ, а также, что является наиболее важным, значительное увеличение амплитуды и частоты фазных сокращений по сравнению с параметрами ЛУ крыс из группы ВЖР (см. рис. 3). Эти данные доказывают, что в брыжеечных ЛУ крыс при ожирении развивается воспаление, оказывающее негативное влияние на сократительную функцию ЛУ. В процессе исследования у части узлов мы удаляли PNAT. Поскольку ЛУ крыс из группы ВЖР с удаленной PNAT демонстрировали достоверно лучшие показатели сократительной функции по сравнению с ЛУ крыс из группы ВЖР с сохраненной PNAT, но достоверно худшие по сравнению с ЛУ крыс контрольной группы (см. рис. 3), мы полагаем, что при ожирении воспаление развивается как в окружающей PNAT, так и непосредственно в тканях ЛУ. Медиаторы воспаления, образующиеся при ожирении в PNAT и диффундирующие в ЛУ, оказывают ингибирующий эффект на сократимость ГМК их капсулы.

Широкий спектр медиаторов воспаления, воздействующих на ЛС, приводит к образованию очень сложной сети сигналов, которая до сих пор полностью не изучена. При этом имеются данные, что при воспалении наблюдается высокая экспрессия iNOS, приводящая к образованию больших количеств NO, оказывающего ингибирующий эффект на сократимость и активную насосную функцию ЛС [24]. Недавно в нашей лаборатории также было показано, что воспаление в брюшной полости крыс сопровождается экспрессией iNOS в брыжеечных ЛУ и ослаблением их сократимости [25]. Для подтверждения участия iNOS в ингибировании сократительной функции ЛУ при ожирении в данном исследовании мы применяли селективный ингибитор iNOS - 1400W. Действие 1400W сопровождалось выраженным повышением тонуса и увеличением амплитуды фазных сокращений ЛУ крыс группы ВЖР и частоты фазных сокращений ЛУ у крыс всех групп, за исключением контрольной и устойчивой к ожирению. Значительные различия амплитуды и частоты спонтанных фазных сокращений в PSS и при действии 1400W позволяют заключить, что так же, как и в ЛС, ожирение способствует экспрессии iNOS в ЛУ и образовании ею значительных количеств NO и что этот механизм является важнейшим, если не основным, приводящим к ингибированию сократительной функции ЛУ при ожирении.

NO в высокой концентрации у животных с ожирением может регулировать функцию ЛС и ЛУ посредством нитрозативного стресса и образования свободных радикалов кислорода (АФК), а клетки ЛС чувствительны к повреждению свободными радикалами [26]. В свою очередь, диета с высоким содержанием жиров провоцирует развитие воспаления в кишечнике, а воспаление сопровождается повышением уровня АФК [27]. Ранее ингибирующий эффект пероксида водорода на сократимость ЛС был продемонстрирован D.C. Zawieja и K.L. Davis на анестезированных крысах in vivo; в их исследовании под влиянием пероксида водорода частота сокращений ЛС снизилась на 85%, а объем прокачиваемой лимфы - на 93% [28]. Примерно такие же результаты были получены ими при добавлении в раствор гипоксантина и ксантиноксидазы. С учетом этих данных в отдельной серии экспериментов мы оценили возможную роль АФК в релаксации ЛУ при ожирении. В нашей работе мы не определяли уровень АФК, а использовали поглотитель АФК - темпол. Через 20 мин после введения в раствор темпола в ЛУ крыс из группы ВЖР достоверно повышались тонус и амплитуда фазных сокращений. Наиболее выраженным был эффект темпола на частоту фазных сокращений ЛУ (повышение на 80%). Темпол также достоверно увеличивал этот показатель в ЛУ крыс из группы ВЖР+ОР. Данные, полученные в этой серии экспериментов, дают основания полагать, что под влиянием ожирения в тканях ЛУ и PNAT образуется значительное количество АФК, которые ингибируют сократительную функцию ГМК капсулы ЛУ.

Ранее было показано, что при воспалении (экспериментальный колит) ЛС мышей способны продуцировать метаболиты арахидоновой кислоты, которые влияют на их сократимость аутокринным образом [29]. Авторы, используя различные ингибиторы, показали, что воспаление приводит к экспрессии в ЛС ЦОГ-2 и повышению содержания простагландина PGE2, который ингибирует сократимость ЛС. В нашей работе с целью выяснения роли простагландинов в релаксации ЛУ мы применяли селективный ингибитор ЦОГ-2 - династат. Применение династата сопровождалось возрастанием амплитуды и частоты фазных сокращений ЛУ крыс из группы ВЖР, что позволило нам сделать заключение об участии простагландинов в релаксации ЛУ при ожирении, хотя их релаксирующий эффект был выражен значительно слабее по сравнению с NO.

В заключительной части нашего исследования мы оценили массу тела, массу висцерального жира и параметры сократительной функции брыжеечных ЛУ крыс с ожирением, которые после потребления ВЖР были переведены на рацион с ограниченной калорийностью. Масса тела этих животных за 8 нед содержания на ограниченном рационе снизилась на 11%, а масса висцерального жира - на 30%. Сопоставление параметров сократительной функции ЛУ крыс из группы ВЖР и ВЖР+ОР показало достоверные различия тонуса (см. рис. 2), а также амплитуды и частоты спонтанных фазных сокращений ЛУ. У крыс после ограничения рациона (ВЖР+ОР) амплитуда фазных сокращений ЛУ была на 22%, а частота на 128% больше по сравнению с ЛУ крыс, получавших ВЖР. Ранее в исследовании на мышах также было показано, что перевод мышей с ожирением на обычную диету приводит к снижению массы тела и нормализации скорости прокачивания лимфы по ЛС [30]. Полученные данные позволяют сделать заключение о том, что перевод крыс с ожирением на рацион с ограниченной калорийностью способствует не только уменьшению массы тела и массы висцерального жира, но и улучшению транспортной функции ЛУ.

Заключение

Ожирение у крыс приводит к нарушению сократительной функции ЛУ (снижаются тонус ЛУ, амплитуда и частота спонтанных фазных сокращений), что может создавать дополнительные препятствия на пути движения лимфы и способствовать просачиванию ее из ЛУ в окружающую жировую ткань.

Ожирение сопровождается развитием воспаления в ЛУ и перинодальной жировой ткани, что приводит к экспрессии индуцибельной NO-синтазы, ЦОГ-2 и накоплению АФК. NO, простагландины и АФК, образующиеся при ожирении, оказывают ингибирующее действие на ГМК капсулы ЛУ, приводя к снижению тонического напряжения и ослаблению фазных сокращений.

Ингибирование сократительной функции ЛУ происходит именно по причине ожирения, поскольку у крыс, получавших ВЖР и устойчивых к ожирению, сократительная функция ЛУ изменялась слабо.

Перевод крыс на диету с ограничением калорийности до 80% от рекомендуемого стандартного рациона сопровождается снижением массы тела, массы висцерального жира и улучшением сократительной функции ЛУ. Эти результаты крайне важны, поскольку они предполагают, что дисфункция ЛУ, вызванная ожирением, является обратимой (по крайней мере частично).
