Effect of low doses of active chlorine on commensal populations of intestinal bacteria in rats by oral ingestion

Abstract

Chlorine-containing substances are widely used as disinfectants for treating equipment surfaces and technological aids for antimicrobial treatment of a number of foodstuff in the food industry. The toxic and bactericidal effects of the active (free) chlorine they contain are well understood for the concentrations used in practice, whereas little is known about the effect of its residual (subinhibitory) amounts on the organism and on the microbiota, including the ability to induce antimicrobial resistance.

The aim of the study was to investigate the effect of different doses of active chlorine at oral administration on the commensal bacteria of intestinal microbiota, body weight gain and micromorphological features of the liver in rats.

Material and methods. The study was carried out on male Wistar rats, with an initial body weight of 90–100 g, which for 4 weeks received doses of active chlorine together with drinking water ad libitum, with the given concentration: subinhibitory (10 mg/L), threshold (50 mg/L) and aggravated (100 mg/L). Chloramine was used as a chlorine-containing agent stable in aqueous solution. Body weight gain was monitored daily. After decapitation, the caecum was taken for microbiota examination as well as the liver. The phenotypic antimicrobial resistance characteristics of Enterobacteriaceae and Enterococci were studied by the disk diffusion method. Additionally, a micromorphologic study of liver slices was performed.

Results. Insignificant negative deviations in the body weight gain of rats in the experimental groups receiving subinhibitory doses of active chlorine, combined with a reduced level of Enterococci and Enterobacteriaceae representatives, were revealed. No significant effect of chlorine on the levels of resistant Escherichia coli populations was found, but a tendency to exhibit antimicrobial resistance of Enterococci isolated from rats receiving low doses of active chlorine (10 and 50 mg/l) was detected. No signs of toxic effect on the liver tissue most sensitive to chlorine were detected, but some accumulations of inflammatory cells in the liver slice were revealed.

Conclusion. Low doses of chlorine-containing substances at their oral consumption are not indifferent for rats’ organism, causing negative phenomena in intestinal bacteria and in liver tissues at the level of tendency. It is expedient to continue studies in this direction.

В табл. 5 представлены характеристики фенотипических профилей АМР штаммов E. coli, выделенных из содержимого толстой кишки крыс (25 культур).

Установлено, что профили чувствительности штаммов E. coli от крыс основных групп не отличались существенно от таковых у животных контрольной группы. Все изученные изоляты энтеробактерий были чувствительны к аминогликозидам (амикацину, гентамицину), к фторхинолонам (налидиксовой кислоте, ципрофлоксацину, левофлоксацину) и хлорамфениколу. Резистентность к нитрофурантоину обнаружена у 1 штамма E. coli (4%). Исследованная популяция штаммов характеризовалась высокой распространенностью резистентности к антибиотикам тетрациклиновой группы. Это позволяет сделать вывод, что все испытанные дозы хлора не оказали влияния на профиль резистентности кишечных бактерий к антимикробным препаратам в сравнении с контролем.

Результаты определения чувствительности энтерококков, выделенных из содержимого кишечника крыс основных и контрольной групп, к ампициллину, ванкомицину, эритромицину, ципрофлоксацину, левофлоксацину, тетрациклинам, хлорамфениколу, нитрофурантоину и линезолиду приведены в табл. 6.

Как следует из полученных данных, профиль чувствительности штаммов энтерококков от крыс, получавших растворы хлора, так же как и в случае E. coli, не различался значимо от контроля. Среди них не было штаммов, устойчивых к антимикробному препарату резерва имипенему. Но в целом в изученной выборке выявлялись штаммы, устойчивые к ампициллину, ципрофлоксацину, линезолиду и тетрациклинам. В то же время из групп, получавших низкие дозы активного хлора, были выделены штаммы E. faecium, устойчивые сразу к нескольким антимикробным препаратам, в разных комбинациях. Так, штамм E. faecium № 84хэ проявлял резистентность одновременно к 8 антимикробным препаратам (тетрациклинам, ампициллину, ципрофлоксацину, ванкомицину, эритромицину и линезолиду), E. faecium № 15хэ - к 7 антибиотикам. Полученные данные подтверждают, что антибиотикорезистентные фенотипы энтерококков присущи в основном виду E. faecium, обладающему повышенной способностью к горизонтальному трансферу генов АМР [20, 21].

Сопоставление профилей АМР энтерококков, выделенных от животных из основных и контрольной групп, свидетельствует о большой частоте признаков фенотипической резистентности в группах, получавших растворы с концентрацией активного хлора 10 и 50 мг/л - 32 и 38% случаев выявления резистентности, тогда как в группе, получавшей 100 мг/л хлора, и в контроле - соответственно 18 и 25%.

В целом в данном эксперименте in vivo не выявлено влияния водных растворов хлора на приобретение АМР у штаммов E. coli, выделенных из кишечника крыс; полученные данные показывают тенденцию формирования корезистентности к антимикробным препаратам у выделенных штаммов энтерококков. В исследовании [22] in vitro показано влияние доз 10-100 мг/л активного хлора на частоту конъюгативного переноса мобильных генетических элементов от E. coli к Salmonella typhimurium, при этом концентрации хлора, превышающие 100 мг/л, способствовали ингибиции роста исследуемых видов бактерий и подавлению их способности к фенотипической АМР, так же как и в проведенном нами исследовании. По-видимому, для обнаружения более явных эффектов формирования АМР целесообразно проведение пролонгированных исследований in vivo и с оценкой большего объема выборки штаммов3.

Изучение влияния низких доз активного хлора на печень крыс

Дополнительные возможные негативные эффекты низких доз хлора были также оценены в микроморфологическом (гистологическом) исследовании срезов печени как органа, который участвует в детоксикации ксенобиотика в организме.

Известны работы, в которых применялся аналитический показатель для оценки токсического воздействия соединений хлора [23]. Но выявленные различия носили характер тенденции.

Общая картина гистологического состояния ткани печени у крыс, получавших хлор с водой, представлена в табл. 7. Микроскопическое исследование срезов печени показало, что строение долек и структура печени в целом сохранены. Печеночные балки в виде пластин из гепатоцитов радиально расходятся от периферии к центральной вене, разделены синусоидальными капиллярами, в которых обнаруживаются форменные элементы крови, эритроцитарные массы насыщенно-красного цвета. Кровенаполнение синусоидных капилляров варьирует от слабого кровенаполнения до умеренного их полнокровия. Желчные капилляры обычного размера, внутрипеченочного и внутрипротокового застоя желчи не отмечено, пролиферации желчных протоков также не отмечено.

Гепатоциты нормального размера, имеют полиэдральную форму с расположенным в центре округлым крупным ядром, содержащим 1-2 ядрышка. Так, отмечались изменения, которые не носили дозозависимого характера. Цитоплазма некоторых гепатоцитов образцов печени животных из 3-й и 4-й групп содержит небольшие по размеру вакуоли - признак мелкокапельной жировой дистрофии. Цитоплазма гепатоцитов остальных препаратов однородная, без включений и признаков жировой или гидропической (баллонной) дистрофии. Полиплоидные и многоядерные гепатоциты практически отсутствуют.

В паренхиме дольки по ходу синусоидных капилляров диффузно в небольшом количестве расположены мононуклеарные клетки воспалительной инфильтрации. На препаратах, полученных от животных 1-й и 2-й групп, отмечены очаги из скопления воспалительных клеток, организованные как микрогранулемы. Частота этих отклонений составила 17%.

Портальные тракты не расширены, в строме ряда из них очаговая умеренная лимфогистиоцитарная инфильтрация, без признаков склероза.

Таким образом, структурные изменения в ткани печени в исследовании перорального потребления растворов хлора не выявлены, но были отмечены отдельные скопления воспалительных клеток у крыс, получавших субингибиторную и пороговую дозы хлора.

Заключение

Низкие дозы активного хлора, равнозначные уровням допустимых для контакта с пищевыми продуктами (например, при охлаждении тушек птицы - 10 и 50 мг/л), не оказывали достоверных негативных эффектов на уровни представителей резистентных E. coli в кишечнике крыс, а также на профиль их резистентности к наиболее распространенным антимикробным препаратам. При этом отмечено, что почти все штаммы E. faecium в группе животных, получавших низкие дозы активного хлора, имели устойчивость сразу к нескольким антибиотикам; среди этих энтерококков выявлены 2 мультирезистентных штамма E. faecium, устойчивых одновременно к 7-8 антибиотикам. Сопоставление профилей АМР выявленных штаммов в группах, получавших разные дозы активного хлора, свидетельствовало о тенденции развития устойчивости на дозах 10 и 50 мг/л активного хлора, но не на дозе 100 мг/л.

Пероральное поступление субингибиторных доз активного хлора не может быть индифферентным для организма, поскольку темпы прироста массы тела крыс, получавших 10 мг/л хлора, на 4-й неделе были ниже в сравнении с контролем или совпадали с тенденцией снижения относительной массы печени у крыс, получавших субингибиторную и пороговую дозы, с выявлением отдельных скоплений воспалительных клеток (на фоне отсутствия морфологических изменений в ткани печени), а также с тенденцией к снижению уровня энтеробактерий и энтерококков в кишечной микрофлоре. Это обусловливает необходимость продолжения исследований в данном направлении, в том числе в отношении пересмотра NOAEL (максимально недействующей дозы) хлора на микробиоту.
