Development of fermented milk product based on mare milk and lactic microorganisms association

Abstract

Since ancient times, mare’s milk has been widely used by many peoples. Thanks to its unique composition, it is now used in the nutrition of people with allergies to cow’s milk, in the technologies of foods for special dietary uses, etc. To expand the range of products with useful properties, it is advisable to develop fermented milk products based on mare’s milk. Numerous studies indicate that the use of fermented milk products provides various health benefits. And the use of probiotic cultures in the composition of starter cultures for fermented milk products allows you to add a number of functional properties to the product. However, the only fermented milk product available on the market from mare’s milk is koumiss. Therefore, the development of new fermented milk products based on mare’s milk is relevant and in demand.

The aim of the study was to develop a fermented milk product based on mare’s milk using an association consisting of yogurt starter culture and a probiotic strain of L. rhamnosus F.

Material and methods. For the development of a fermented milk product based on mare’s milk, the yogurt starter STBp (S. thermophilus and L. delbrueckii subsp bulgaricus) and the probiotic culture L. rhamnosus F (GenBank MN994629) from the collection of lactic acid and probiotic microorganisms of VNIMI were chosen. As a basis for the fermented milk product, mare’s milk was used with the addition of dry mare’s or dry cow’s milk, pre-pasteurized at a temperature of 65±1 °C with a holding time of 30 min and cooled to a fermentation temperature of 37±1 °C. In the course of the work, the activity of acid formation during product fermentation, the duration of fermentation and the dynamics of the number of lactic acid bacteria and L. rhamnosus F probiotic strain during fermentation were studied, depending on the ratio of cultures in the ferment, the dose of the ferment introduced and the base for fermentation. The antimicrobial activity of the product was determined by agar well diffusion assay.

Results. It was found that L. rhamnosus F has a low acid-forming activity, so the use of a combined starter culture (association) was proposed. It was determined that the addition of cow’s milk powder to mare’s milk positively affects the activity of acid formation; after 6 hours of fermentation, the pH value varied in the range of 4.6-4.83, and after 8 hours pH was 4.44-4.65. When dry mare’s milk was added, the pH value after 8 hours of fermentation was in the range of 4.71-4.98 pH units. The influence of the amount of combined starter culture (association) and its cultures ratio, the duration of fermentation on the content of lactic acid bacteria, including the probiotic strain L. rhamnosus F in the fermented milk product based on mare’s milk, was established. The largest amount of L. rhamnosus F was contained in the product fermented by 7% association with the ratio of cultures 1/4 and 1/6 and after 8 h it amounted to 7-9.5x108 CFU/cm3.

Conclusion. Summarizing the results of the research, the technology of a fermented milk product based on mare’s milk was developed: the amount of starter culture (associations) - 7%, the ratio of cultures - 1 part of the starter culture for yogurt and 6 parts of L. rhamnosus F (1/6), the fermentation temperature - 37±1 °C, the duration of fermentation - 6 hours with the addition of cow’s milk powder and 8 hours with the addition of mare’s milk powder. The developed fermented milk product has antimicrobial activity against opportunistic and pathogenic microorganisms E. coli ATCC 25922, S. aureus ATCC 6538, S. typhimurium ATCC 14028.

В процессе сквашивания происходило нарастание титруемой кислотности, которое зависит от основы для сквашивания, количества вносимой закваски (ассоциации), а также от соотношения штаммов в ней. Чем больше в комбинированной закваске L. rhamnosus F, тем меньше скорость увеличения титруемой кислотности, т.е. менее интенсивно снижалось содержание лактозы в продукте.

Также в процессе сквашивания определяли количество клеток S. thermophilus и L. bulgaricus, входящих в состав закваски для йогурта, и пробиотического штамма L. rhamnosus F в зависимости от количества закваски (ассоциации), продолжительности сквашивания, соотношения закваски для йогурта и L. rhamnosus F, основы для сквашивания. Данные по изменению количества клеток L. rhamnosus F в процессе сквашивания представлены на рис. 5, 6.

При соотношении культур 1/4 и 1/6 количество клеток L. rhamnosus F в процессе сквашивания молока кобыльего с добавлением сухого кобыльего молока увеличивалось по сравнению с соотношением 1/2. Так, при внесении 5% закваски с соотношением штаммов 1/2 количество клеток L. rhamnosus F через 8 ч сквашивания достигало 9х107 КОЕ/см3, а при соотношении штаммов 1/6 - 3х108 КОЕ/см3. При сквашивании кобыльего молока с добавлением сухого кобыльего молока наибольшее количество пробиотической культуры L. rhamnosus F содержалось в продукте при внесении 7% ассоциации с соотношением культур 1/4 и 1/6 через 8 ч сквашивания и составило 7х108 КОЕ/см3.

Та же тенденция прослеживалась при сквашивании молока кобыльего с добавлением сухого коровьего молока. При использовании для сквашивания закваски в количестве 5% с соотношением штаммов 1/2 количество клеток L. rhamnosus F через 8 ч сквашивания достигало 1,2х108 КОЕ/см3, а при соотношении штаммов 1/6 - 4,2х108 КОЕ/см3. Наибольшее количество клеток L. rhamnosus F при сквашивании кобыльего молока с добавлением сухого коровьего молока содержалось в кисломолочном продукте через 8 ч при внесении 7% закваски (ассоциации) с соотношением культур 1/6 (9,5х108 КОЕ/см3).

Анализ полученных данных по накоплению клеток L. rhamnosus F в процессе сквашивания кобыльего молока с добавлением сухого кобыльего молока и кобыльего молока с добавлением сухого коровьего молока показал, что статистически значимых (p<0,05) различий между выборками не было. При внесении 7% закваски с соотношением культур 1/6 через 8 ч сквашивания количество клеток L. rhamnosus F варьировало незначительно и составляло 7х108 КОЕ/см3 при сквашивании кобыльего молока с добавлением сухого кобыльего молока и 9,5х108 КОЕ/см3 при сквашивании кобыльего молока с добавлением сухого коровьего молока. Полученные результаты показывают, что для технологии кисломолочного продукта в качестве основы для сквашивания может быть использовано кобылье молоко с добавлением сухого кобыльего или сухого коровьего молока.

На рис. 7 представлены результаты исследований по содержанию клеток S. thermophilus + L. bulgaricus, входящих в состав закваски для йогурта, в кисломолочном продукте в зависимости от количества закваски (ассоциации) с соотношением 1/6 (1 часть закваски для йогурта и 6 частей L. rhamnosus F), продолжительности сквашивания и основы для сквашивания.

Содержание клеток (S. thermophilus + L. bulgaricus) при внесении 5% закваски (ассоциации) с соотношением культур 1/6 при использовании кобыльего молока с добавлением сухого коровьего молока уже через 6 ч сквашивания составило 6х108 КОЕ/см3, а при сквашивании кобыльего молока с добавлением сухого кобыльего молока - 2,5х107 КОЕ/см3. Независимо от количества внесенной закваски через 8 ч сквашивания кобыльего молока с добавлением сухого кобыльего молока или сухого коровьего молока количество клеток (S. thermophilus + L. bulgaricus) составило 7х108 КОЕ/см3.

По совокупности полученных данных были разработаны технологические параметры сквашивания кисломолочного продукта на основе кобыльего молока с добавлением сухого кобыльего и сухого коровьего молока с использованием пробиотического штамма L. rhamnosus F и закваски для йогурта: температура сквашивания - 37±1 °С, количество закваски - 7%, соотношение культур в закваске - 1 часть закваски для йогурта и 6 частей L. rhamnosus F (1/6). Продолжительность сквашивания кобыльего молока с добавлением сухого кобыльего молока составляет 8 ч, при этом количество L. rhamnosus F достигает 7х108 КОЕ/см3, а количество микроорганизмов закваски для йогурта - 7х108 КОЕ/см3 Продолжительность сквашивания кобыльего молока с добавлением сухого коровьего молока составляет 6 ч, при этом количество L. rhamnosus F достигает 4,2х108 КОЕ/см3, а количество микроорганизмов закваски для йогурта - 6х108 КОЕ/см3.

Антимикробная активность - одно из важных свойств пробиотических микроорганизмов, используемых при создании кисломолочных продуктов, что позволяет их использовать в разработке функциональных продуктов для улучшения здоровья человека, в связи с этим была определена антимикробная активность разработанного кисломолочного продукта на основе кобыльего молока по отношению к трем условно-патогенным и патогенным микроорганизмам. Данные по антимикробной активности представлены в табл. 2.

В результате проведенных исследований установлено, что разработанный кисломолочный продукт на основе кобыльего молока обладает антимикробной активностью, которая незначительно варьирует в зависимости от выбранных условно-патогенных штаммов.

Заключение

В последние годы заметно вырос интерес к использованию кобыльего молока в технологиях пищевых продуктов для диетического лечебного и профилактического питания, что связано с его составом. Разработка новых кисломолочных продуктов на основе кобыльего молока представляет несомненный интерес для увеличения ассортимента продуктов с полезными свойствами, а использование пробиотических культур в составе закваски позволит добавить продукту ряд функциональных свойств.

Учитывая низкую кислотообразующую активность пробиотического штамма L. rhamnosus F при сквашивании кобыльего молока, для разработки технологии кисломолочного продукта на основе кобыльего молока предложено применение комбинированной закваски (ассоциации), а именно закваски для йогурта и L. rhamnosus F.

В результате проведенных исследований разработана технология кисломолочного продукта на основе кобыльего молока: количество закваски (ассоциации) - 7%, соотношение культур - 1 часть закваски для йогурта и 6 частей L. rhamnosus F (1/6), температура сквашивания - 37±1 °С, продолжительность сквашивания - 6 ч при добавлении сухого коровьего молока и 8 ч при добавлении сухого кобыльего молока. Количество молочнокислых бактерий в продукте - не менее 1х107 КОЕ/см3 и содержание пробиотического штамма L. rhamnosus F не ниже 106 КОЕ/см3.

Разработанный кисломолочный продукт обладает пробиотическими свойствами, а именно антимикробной активностью по отношению к условно-патогенным и патогенным микроорганизмам: E. coli АТСС 25922, S. aureus АТСС 6538, Salmonella typhimurium АТСС 14028.
