Protective effect of anthocyanins on apoptosis of gastrocnemius muscle myocytes of rats after intense exercise

Abstract

Currently, in sports medicine, much attention is paid to the prevention and treatment of delayed muscle soreness syndrome (DOMS), which occurs several hours or days after unusual or intense physical activity, as well as the state of athlete overtraining. One of the main pathogenetic factors in the development of this syndrome is myocyte ultrastructural damage with apoptosis activation. Therefore, using natural antioxidants in sports nutrition for the relief of this pathology is of particular relevance.

The aim of the study was to study the effect of an anthocyanin-enriched diet on apoptosis of gastrocnemius muscle myocytes of rats after intense exercise.

Material and methods. The experiment was carried out for 4 weeks on 4 groups of male Wistar rats (12 animals in each, initial body weight ≈300 g). Animals were divided into groups of rats (groups 1 and 2), whose motor activity was limited by standard conditions for keeping animals in vivarium, and groups of physically active rats (groups 3 and 4), which received additional physical activity – treadmill training. Before the end of the experiment, the animals of groups 3 and 4 were given debilitating (until the rats refused to continue the exercise) physical activity on a treadmill. Rats of all four groups received a standard semi-synthetic diet, water ad libitum. Animals in groups 2 and 4 were additionally given blueberry and blackcurrant extract (30% anthocyanins) as part of the diet at a daily dose of 15 mg anthocyanins/kg body weight. The intensity of apoptosis of gastrocnemius muscle myocytes was studied by flow cytometry. Cells were stained with fluorochrome-conjugated annexin V and vital dye 7-aminoactinomycin. The results are presented as the percentage of intact cells and cells at different stages of apoptosis per 100 000 counted objects in each sample.

Results. The enrichment of the diet of control group rats with blueberry and black currant extract did not have a significant effect on the relative content of intact cells and the studied parameters of apoptosis of gastrocnemius muscle myocytes of rats of the 2nd group. Intense physical activity in rats of the 3rd group led to a statistically significant (p<0.05) decrease in the relative content of intact (live) cells compared with this indicator in rats of other groups (85.32±1.44 vs 90.87±0.66% – in the 1st group; 90.16±0.79% – in the 2nd group; 89.01±0.81% – in the 4th group, р<0.05). After intense physical activity in rats of the 3rd group, activation of apoptosis of gastrocnemius muscle myocytes was found, as evidenced by an increase in the relative content of objects in apoptosis compared with other groups (11.61±1.45 vs 7.88±0.60% – in the 1st group, р<0.05; 8.01±0.70% – in the 2nd group, p<0.10; 7.93±0.59% – in the 4th group р<0.05). Enrichment of the diet of exercise rats with blueberry and blackcurrant extract (4th group) had a protective effect on the intensity of the apoptosis process, the studied parameters did not differ significantly from those in rats of the control and the 2nd groups.

Conclusion. The results of the study indicate the activation of the process of apoptosis of gastrocnemius muscle myocytes of rats after intense physical activity. Enrichment of rats’ diet with anthocyanins from blueberry and black currant extracts ensures the restoration of the studied apoptosis parameters to the level of control group rats. In the control group of rats with normal physical activity, the addition of anthocyanins to the diet does not have a significant effect on the physiological process of apoptosis of gastrocnemius muscle myocytes. In this way, an evidence base for the effectiveness of the use of biologically active substances – anthocyanins – in sports nutrition for the restoration of skeletal muscles has been obtained.

Апоптоз - физиологический процесс генетически запрограммированной гибели клетки с характерными морфологическими и биохимическими признаками [1]. Он обеспечивает тканевый гомеостаз и регулирует объем тканей, уравновешивая их новообразование. Апоптоз, вызванный физическими упражнениями, является нормальным регуляторным процессом, который служит для удаления поврежденных клеток без выраженной воспалительной реакции, обеспечивая тем самым оптимальное функционирование организма [2]. Ему подвергаются все элементы скелетных мышц: миофибриллы, миоциты, сателлитные клетки, эндотелиальные клетки [3]. Инициируют апоптоз многие факторы. Сигналы к его запуску могут быть рецепторными или внутриклеточными (нерецепторными). Нерецепторными сигналами к апоптозу являются изменение потенциала и дестабилизация мембраны митохондрий с помощью проапоптогенных белков семейства Вcl-2 (Bax, Bcl-xs, Bid и др.) или белка р53, который контролирует клеточный цикл и активируется при его нарушении [4]. Ключевыми факторами, индуцирующими проапоптогенные белки, являются изменение оксидантного статуса клетки, образование активных форм кислорода и нарушение процессов активации клетки [5], что может быть следствием, в частности, интенсивных физических нагрузок и состояния перетренированности [6]. После интенсивных физических нагрузок у спортсменов часто развивается синдром отсроченной мышечной боли - это тип ультраструктурного повреждения мышц [7]. Клинические признаки включают снижение силовых способностей, болезненное ограничение движений, скованность, отек и дисфункцию соседних суставов. Хотя эта патология считается легким типом травмы, она является наиболее распространенной причиной снижения спортивных результатов. Одним из основных механизмов патогенеза является апоптоз миоцитов скелетной мускулатуры.

Поскольку основным патогенетическим механизмом в развитии мышечного повреждения является окислительный стресс [8], в последние годы в спортивном питании стали широко применяться антиоксиданты [9-12]. При этом особое внимание уделяется антиоксидантной роли полифенолов, полученных из фруктов, применение которых обеспечивает оптимальный уровень восстановления организма после физических нагрузок и ряда неинфекционных заболеваний [13]. В рационы спортсменов включали различные фрукты, например новозеландскую черную смородину, гранат и вишню в виде экстрактов (многокомпонентных или очищенных), соков и настоек. Полифенолы широко представлены в пищевой продукции растительного происхождения. Антоцианины, водорастворимый подкласс флавоноидов, являясь естественными пигментами, придающими пурпурную, синюю, красную и оранжевую окраску цветам, листьям, фруктам и овощам, обладают вазопротекторными свойствами, оказывают антиоксидантное, противовоспалительное, антиатерогенное и сосудорасширяющее действие [14-16]. Для взрослых адекватный уровень потребления антоцианинов, согласно МР 2.3.1.0253-21 "Нормы физиологических потребностей в энергии и пищевых веществах для различных групп населения Российской Федерации", составляет 50 мг/сут [17]. Согласно [18], анализ онлайн-баз данных MEDLINE, ScienceDirect, Cochrane Librare и SPORTDiscus показал, что в спортивной медицине используются дозировки антоцианинов от 18 до 552 мг/сут.

Цель исследования - изучение влияния обогащения рациона антоцианинами на апоптоз миоцитов икроножной мышцы крыс после интенсивной физической нагрузки.

Материал и методы

Эксперимент проводили в течение 4 нед на 4 группах крыс-самцов линии Wistar (по 12 животных, исходная масса тела ≈300 г), полученных из питомника филиала "Столбовая" ФГБУН "Научный центр биомедицинских технологий ФМБА". Исследование получило одобрение этического комитета ФГБУН "ФИЦ питания и биотехнологии" (заседание № 11 от 15.12.2021) и проводилось в соответствии с ГОСТ 33216-2014 "Руководство по содержанию и уходу за лабораторными животными. Правила содержания и ухода за лабораторными грызунами и кроликами". Животных содержали по 2 особи в пластиковых клетках на подстилке из древесных стружек при искусственном освещении с равной продолжительностью ночного и дневного периодов.

В рамках эксперимента животных разделяли на группы крыс (1-я и 2-я группы), двигательная активность которых ограничивалась стандартными условиями содержания животных в виварии, и группы физически активных крыс (3-я и 4-я группы), которые получали дополнительную физическую нагрузку - занятия на беговой дорожке (3 раза в неделю, угол 10°, скорость 15 м/мин, продолжительность 20 мин). Непосредственно перед выведением из эксперимента животным 3-й и 4-й групп давали истощающую (до отказа крыс от продолжения упражнения) физическую нагрузку на беговой дорожке (угол 10°, скорость 12 м/мин в течение 3 мин, далее повышение скорости на 1,2 м/мин каждые 30 с до скорости 38,4 м/мин).

Крысы всех 4 групп получали стандартный полусинтетический рацион на основе АIN 93M [19] из расчета 25 г/сут сухого корма на крысу, воду ad libitum. Животные 2-й и 4-й групп в составе рациона дополнительно получали экстракт черники и черной смородины (30% антоцианинов, Healthberry 865, Evonik Nutrition & Care GmbH, Германия) [20] в суточной дозе 15 мг антоцианинов на 1 кг массы тела. Отъем корма проводили за 16 ч до выведения животных из эксперимента.

После декапитации под эфирным наркозом у животных выделяли из икроножной мышцы образец мышечной ткани для дальнейшего изучения.

Интенсивность апоптоза миоцитов икроножной мышцы оценивали методом проточной цитометрии. Суспензию клеток получали с помощью автоматической системы для механической гомогенизации ткани (BD Medimachine, Becton Dickenson and Company, США). Однократно отмывали клетки забуференным фосфатами 0,15 М раствором хлорида натрия, рН 7,2-7,4 и готовили пробу с концентрацией клеток 1×106/см3. Окрашивание клеток проводили конъюгированным с флуорохромом аннексином V и витальным красителем 7-аминоактиномицином (7-AAD) (Beckman Coulter, США) с последующей детекцией на проточном цитофлуориметре (FC-500, Beckman Coulter, США). Иссеченные образцы и клеточная суспензия в процессе работы хранились на льду. Результаты представлены в виде процентного соотношения интактных клеток и клеток, находящихся на разных стадиях апоптоза на 100 000 просчитанных объектов в каждом образце. Принцип метода основан на свойстве аннексина V связываться с фосфатидилсерином мембраны и способности 7-AAD встраиваться между цитозином и гуанином двухцепочечной ДНК клеток с нарушенной целостностью мембраны. На ранних стадиях апоптоза целостность клеточной мембраны сохраняется, но происходит конверсия мембранных фосфолипидов и появление фосфатидилсерина на поверхности клетки. Аннексин V с высокой аффинностью связывается с фосфатидилсерином и идентифицирует "ранний" апоптоз, т.е. AnV-FITC+/7-AAD-. Поздняя фаза апоптоза сопровождается не только формированием мембранной асимметрии, но и нарушением целостности мембраны, фрагментацией ДНК и резким возрастанием мембранной проницаемости для катионных красителей. Такая стадия апоптоза детектируется как AnV-FITC+/7-AAD+. Мертвые клетки выявляются как AnV-FITC-/7-AAD+. Интактные клетки не связывают AnV-FITC и непроницаемы для 7-AAD, т.е. AnV-FITC-/7-AAD-.

Статистический анализ данных выполняли с использованием 2-факторного дисперсионного анализа ANOVA. Гипотезу о различии функции распределения данных в сравниваемых группах дополнительно проверяли с использованием непараметрического критерия Манна-Уитни. Различия принимали за достоверные на уровне значимости p<0,05. Расчеты выполняли в пакете программ SPSS 20.0 (IBM, США).
