Biological activity and prospects for food application of cyclic peptides of flax (Linum usitatissimum)
AbstractNowadays all over the world there is an active search for new promising bioactive compounds and evaluation of their efficiency and safety for the usage in food for special dietary uses in order to form balanced healthy diets for different groups of consumers (children, pregnant and lactating women, etc.) or inclusion in diet therapy in order to increase the effectiveness of prevention and treatment of non-communicable diseases and reduce the need for medical care. Such substances can be cyclic peptides – compounds with a unique structure, thanks to which the stability of their configuration and high bioavailability are achieved, which, in turn, allows them to exhibit a wide range of biological effects. Basically, these bioactive compounds are extracted from the leaves and stems of medicinal plants or plants that have no tradition of food use in our country. Common flax (Linum usitatissimum), a crop widely used in nutrition, is practically their only food source.
The purpose of the research was to generalize information on biological activity of flax cyclic peptides in order to assess the prospects of their food application.
Material and methods. Systematic analysis of scientific literature was carried out using keywords (Linseed, or Flaxseed, or Flax, or Linum usitatissimum) and (Orbitide, or Cyclolinopeptide, or Linusorb, or LOMIX) in Google Scholar, PubMed systems. A total of 500 publications were identified and analyzed. Publish or Perish software was used for analysis. The topics of the publications were analyzed using KH Coder software.
Results. Cyclic peptides are among the least studied components of flax, and the profile of cyclic peptides of domestic flax seed varieties has not been described so far. At the same time, the results of preclinical studies of flax cyclic peptides represent promising data that open prospects for their use in the therapy of various diseases. Immunosuppressive and anti-inflammatory activity, as well as hypocholesterolemic and antioxidant effects of flax cyclic peptides have been demonstrated.
Conclusion. The current information on the biological activity of cyclic peptides is incomplete and should be expanded by new data obtained, in particular, in in vivo models and clinical trials. The study of the composition of flax cyclic peptides from domestic raw materials, the development and implementation of the methods of their extraction will contribute to the sustainable development of the industry and will allow reorienting the production of oilseed raw materials from its export to the introduction of knowledge-intensive technologies.

Улучшение качества и продолжительности жизни населения, в том числе путем эффективной профилактики алиментарно-зависимых заболеваний на основе принципов здорового питания, предусматривающих использование различных биологически активных веществ (БАВ), является глобальной проблемой, на решение которой нацелено профессиональное сообщество ученых и практиков. В настоящее время спектр этих веществ является значительным, однако практическое использование в составах пищевых продуктов распространяется на ограниченный перечень, включающий витамины, минеральные вещества, ω-3 полиненасыщенные жирные кислоты, пищевые волокна и пробиотические микроорганизмы. Это связано, в частности, с отсутствием доступных технологий получения БАВ в виде функциональных пищевых ингредиентов. В последние годы все большее внимание уделяется минорным компонентам пищи, высокая биологическая активность которых может обеспечить создание новых видов эффективных специализированных пищевых продуктов [1]. К числу таких соединений относятся циклические пептиды растений [2].
Циклические пептиды были идентифицированы в растениях семейств Анноновые (Annonaceae), Гвоздичные (Caryophyllaceae), Молочайные (Euphorbiaceae, например, маниок), Яснотковые (Lamiaceae, лаванда, базилик, мята, розмарин, чабер, шалфей, майоран, мелисса, тимьян и душица), Льновые (Linaceae), Лаконосовые (Phytolaccaceae), Рутовые (Rutaceae, представители рода Цитрус), Лимонниковые (Schizandraceae), Вербеновые (Verbenaceae) [3]. Различные типы циклических пептидов, выделенные из растений, продемонстрировали разнонаправленные биологические эффекты, такие как гипогликемические, антиоксидантные, гипохолестеринемические, антиканцерогенные, кардиозащитные и иммуносупрессивные [4]. Соответственно, представляется актуальным повышение эффективности использования циклических пептидов из растительного сырья, которые традиционно не являются целевым продуктом, но при этом представляют собой ценный компонент с высокой биологической активностью.
Эти БАВ выделяют в основном из листьев и стеблей лекарственных растений или растений, которые не имеют традиций пищевого применения, поэтому лен обыкновенный является практически единственным их источником, представляющим собой одну из наиболее широко используемых в питании культур и к настоящему времени возделываемой во многих странах мира.
Лен обыкновенный, или лен посевной (Linum usitatissimum) - однолетнее травянистое растение, произрастающее на территории Азии, Америки, Европы и даже части Африки. Одной из самых ценных частей льна является его семя за счет высокого содержания в нем макро- и микронутриентов, а также минорных БАВ [5].
В особенности семя льна является богатым источником циклических пептидов, которые представляют собой циклоолигопептиды (8-10 аминокислотных остатков) с низкой молекулярной массой (~1 кДа). Начиная с 1959 г., в льняном масле, корнях и семенах был идентифицирован 31 циклический пептид, еще 8 пептидов было синтезировано на основе природных вариантов для получения пептидов с заданными свойствами [6]. Циклические пептиды льна имеют различные названия: орбитиды, циклолинопептиды, линусорбы. На рис. 1 представлены сведения об основных натуральных циклических пептидах льна согласно номенклатуре [7].

Несмотря на более чем полувековую историю изучения, систематическая номенклатура циклических пептидов льна была предложена сравнительно недавно, в 2014 г. в работе [7] и пока обсуждается комиссией IUPAC [8].
Считается, что циклические пептиды льна синтезируются из 3 основных последовательностей нуклеотидов AFSQ01016651.1 (1, 2, 8 и 19), AFSQ01025165.1 (5, 11 и 15), AFSQ01011783.1 (21) [9]. После синтеза многие орбитиды претерпевают посттрансляционную модификацию в результате окисления метионина в S-оксид метионина, а затем в S,S-диоксид метионина [10]. Анализ генома льна свидетельствует о большем потенциальном разнообразии циклических пептидов льна, которые в настоящее время еще экспериментально не идентифицированы [11].
Приведенная на рис. 1 номенклатура в настоящее время в большей степени позволяет сформировать понимание разнообразия описываемых в научной литературе циклических пептидов. Тем не менее существует расширенная классификация циклических пептидов льна через обозначение названий посредством термина "линусорб" [6, 12]. Обозначение названий циклических пептидов с использованием новой терминологии обычно приводится в дополнение к более традиционному названию.
В России циклическими пептидами занимаются преимущественно в контексте поиска новых антибиотиков (например, циклические липопептиды - эхинокандины [13]) и пищевой токсикологии (например, циклические пептиды ядовитых грибов - аматоксины). Циклические пептиды льна являются его малоизучаемыми компонентами. Профиль циклических пептидов семян льна отечественных сортов до настоящего времени не описан. Оценка белковых компонентов льняного семени чаще всего сводится к выделению белка для пищевых и кормовых целей. Исследования биологической эффективности циклических пептидов льна носят косвенный характер и преимущественно затрагивают их действие только в комплексе с другими компонентами льняного семени.
Цель исследования - обобщение сведений о биологической активности циклических пептидов льна обыкновенного (Linum usitatissimum) для выявления перспектив пищевого применения.
Материал и методы
Для выявления основных направлений исследований был проведен систематический анализ научной литературы по ключевым словам (Linseed, или Flaxseed, или Flax, или Linum usitatissimum) и (Orbitide, или Cyclolinopeptide, или Linusorb, или LOMIX) в системах Google Scholar, PubMed. В общей сложности было выявлено и проанализировано 500 публикаций. Для анализа использовали программное обеспечение Publish or Perish (версия 8) [14]. Анализ тематики публикаций проводили при помощи программы KH Coder (версия 3.Beta.07b) [15, 16] для построения карт совместного употребления слов в текстах публикаций. На карту взаимосвязей были выведены слова, имеющие по меньшей мере 15 упоминаний. Были отображены только 60 наиболее часто используемых терминов.
Результаты
Как показано на рис. 2, основными тематиками исследований в настоящее время являются изучение биологической активности циклических пептидов льна, при этом большинство исследований посвящено изучению их иммуносупрессивной активности и потенциальной цитотоксичности по отношению к раковым клеткам. Кроме того, авторы изучают генетическое разнообразие льна, химический состав его семян, разрабатывают методы экстракции и синтеза циклических пептидов льна.

Профиль авторских коллективов специфичен и характеризуется участием небольшого числа малых и средних экспертных сообществ (рис. 3).

Лидером в области разработки методов экстракции орбитидов льна является канадский коллектив в составе Y.Y. Shim, P.D. Jadhav, M.J.T. Reaney и др. из института Саскачевана (Канада). Коллектив находится в тесном сотрудничестве с производителем продуктов из семян льна (Prairie Tide Diversified Inc.), совместно с которым осуществляет выработку коммерческого препарата смеси из 6 орбитидов льна LOMIX. Вторым по публикационной активности является коллектив Наньчанского Университета (Китай) в составе X.G. Zou, J.Y. Yang, Z.Y. Deng и др. Данный коллектив занимается преимущественно изучением свойств орбитидов in vitro на культурах клеток, в частности на раковых клетках и в условиях индуцированного воспаления, также коллектив занимается разработкой аналитических методов исследования циклических пептидов льна. Указанные коллективы, ведя независимые исследования, тем не менее имеют совместные публикации по различным направлениям работ.
Коллективы исследователей из Польской академии наук, Вроцлавского университета и Лодзинского технического университета специализируются преимущественно на методах химического синтеза орбитидов. Коллектив исследователей из Токийского университета фармацевтики и наук о жизни (Япония) занимается идентификацией и выявлением новых вариантов циклических пептидов льна.
В России существует несколько профильных отраслевых научных институтов, занимающихся селекцией льна. Однако в современных экспериментальных и обзорных работах по положительному действию компонентов семян льна на здоровье человека упоминания о циклических пептидах отсутствуют [5, 17, 18].
До сих пор нет единого представления о биологической роли циклических пептидов в самом растении. Имеются отдельные предположения об их антиоксидантной [19] и фунгицидной роли [20]. При этом в настоящее время описан ряд свойств циклических пептидов льна как экзогенных БАВ.
Биологическая активность
Авторы некоторых исследований, в том числе обзорных работ [21], предполагают, что биологические эффекты циклических пептидов льна аналогичны эффектам цельного льняного масла, действие которого может быть обусловлено их присутствием. В настоящее время ведутся немногочисленные, но разноплановые исследования их биологической эффективности в условиях in vitro и in vivo (см. таблицу).
